The 1600 Ceribian Tour was the 200th edition of Ceribian Tour, the second most important of cycling's Grand Tours after the Tour de Valland. The complete route of the 1581 Ceribian Tour was announced in early Elva.

Pre-race favourites
The main favourite was Mestran Stavros Michelakakis, the winner of the 1581 Tour de Valland who aimed to become the first rider since 1568 to win the Tour de Valland and the Ceribian Tour in the same season. Only four riders had achieved that: Vallish Mathieu Rozenthal, Nerysians Kurt von Althoff and Jan Schrieber and Alstarian Hans Weltsch.
His main rivals were expected to be 1580 winner Miks Lieltirums and 1581 Tour de Valland winner Johan Van Engelen, along a group of contenders including Sainamese Asai Satoshi, Adwestene Marc Menethil, Nylanders Robbie Bristow and Antonio Andersson, Strathaens Kieran McLucais and Kelly McCullain, Mordvanian Slavoj Savincek, Florinthian Jonathan Keyen, Sequoian Christophe Malletze, or Ceribians Murtaz Sukhisvili, Kakha Arsenidze, and Zurab Avakidze.

Classification leadership
In the 1581 Ceribian Tour, four different jerseys were awarded. For the general classification, calculated by adding each cyclist's finishing times on each stage, and allowing time bonuses for the first three finishers on every stage, the leader received a golden jersey. This classification was considered the most important of the Ceribian Tour, and the winner was considered the winner of the Tour.
Additionally, there was a points classification, which awarded a blue jersey. In the points classification, cyclists got points for finishing in the top 10 in a stage. In addition, points could be won in intermediate sprints.
There was also a mountains classification, the leadership of which was marked by a white jersey with blue polka dots. In the mountains classifications, points were won by reaching the top of a climb before other cyclists. Each climb was categorized as either first, second, third, or fourth-category, with more points available for the higher-categorized climbs.
The fourth jersey represented the young rider classification, marked by a white jersey. This was decided the same way as the general classification, but only riders 25 years old or younger were eligible.
There were also the team classification. In the team classification, the times of the best four cyclists per team on each stage were added; the leading team was the team with the lowest total time.
